What is the goal of Lumon and Kier? I didn’t really get it. Was Mark the only one who could work on Gemma’s files? If so, what were the rest of MDR doing?

Kier and Lumen are trying to end pain and suffering. The either was a first attempt, severance is the latest.

No, everyone can do the numbers, but they aren't as good at it as Mark is. Ever since the first episode it has been noted that Mark is particularly good at completing the files.
